About uterine fibroids
Its incidence rate increases with age. About 20% -50% of women of childbearing age are affected by it. Some patients suffer from anemia, infertility or poor quality of life due to serious symptoms.
Targeted hormone pathway:
1.GnRH antagonists (such as Relugolix): rapidly reduce estrogen levels, combined with hormone replacement therapy (HRT) to reduce side effects.2.Selective Progesterone Receptor Modulators (SPRMs): New generation drugs such as Vilaprsan reduce the risk of liver toxicity while reducing the volume of fibroids.
Non hormonal targets:1.TGF - β pathway inhibitor: Targeted regulation of proliferation and fibrosis of myomas.
2.Epigenetic regulators: such as histone deacetylase (HDAC) inhibitors, intervene in the expression of fibroid related genes.3.Gene therapy: RNA interference technology targeting driver genes such as MED12 is still in the preclinical research stage.
Multiple Phase II/III clinical trials have focused on combination therapies (such as Relugolix+estrogen/progesterone) and novel SPRMs, some of which have shown potential for long-term control of fibroid growth.

Compared to commonly used models such as mice and rats, Guinea Pig exhibits the following advantages in uterine fibroid research:The incidence of spontaneous uterine fibroids is relatively high: elderly female guinea pigs are prone to spontaneous formation of fibroids due to changes in hormone levels, which is closer to the natural course of human disease.
Hormone sensitivity: The physiological response of guinea pig uterus to estrogen/progesterone is similar to that of humans, and is suitable for evaluating the efficacy of hormone drugs.High matching degree of pathological features:
1.The histological structure of fibroids is consistent with that of humans (smooth muscle cell proliferation, extracellular matrix deposition);
2.Accompanied by similar hemodynamic changes (such as thickening of uterine arteries).
3.Pharmacokinetic characteristics: The drug metabolism enzyme spectrum of guinea pigs is closer to that of humans, which can improve the accuracy of drug toxicity prediction.KCI has established a mature guinea pig uterine fibroid model, which can be applied in fields such as drug screening, mechanism research, and combination therapy evaluation.
